The TAACF maintains a continuous flow of compounds into the screening system, advancing candidate compounds through higher level assays as dictated by our assay results. Go to a description of our Assays.
Activity at the primary screening level is determined by the percentage of TB growth inhibited by the sample. The H37Rv strain is used for the first level screening. A sample is considered active when the % Inhibition is greater than or equal to 90%.
Once determined as active, samples continue on to the second level of screening. Here the MIC is determined to an exact value. Activity at the secondary level is determined by examining both MIC and cytotoxicity data on the sample. The H37Rv strain is also used in the secondary assays.
At the conclusion of secondary screening, the MIC and IC50 values are formed into a ratio termed Selectivity Index (SI). At an SI level of 10 or greater, a compound is considered active at the second level.
