The Tuberculosis Antimicrobial Acquisition and Coordinating Facility (TAACF) was established in 1994 by the National Institute of Allergy and Infectious Diseases to facilitate the discovery of new antitubercular drugs.  The program, managed by Southern Research Institute, assists commercial, academic and government laboratories worldwide in identifying new chemical classes of compounds for evaluation in in vitro and in vivo models.  These screening activities have resulted in several promising agents that have reached advanced stages of testing.

The TAACF recently refocused its efforts following a review by a panel of academic and industry experts in anti-infective drug discovery and development.  The TAACF now provides rapid turnaround of data collection to support suppliers with dedicated programs for the discovery of new anti-tubercular agents and who can commit to providing sufficient compounds (at least 1.0 gram) for advanced testing in vivo.  Suppliers who cannot provide scale up for advanced testing but who have access to novel chemical structures may be assisted provided that testing data will be made publicly available.  Priority status is given to compounds with activity on drug resistant strains, and compounds with the potential to shorten therapy by being active on non-replicating persistent organisms.  A new 384-well format was developed for initial screening and MIC testing and was recently used to screen a library of 200,000 compounds.  MIC and cytotoxicity testing of hits is now in progress and the data will be made publicly available later this year.  Active compounds in the Prestwick library of marketed drugs were identified and several are being further evaluated.  TAACF medicinal chemists work closely with suppliers wishing to submit new agents for biological evaluation to identify appropriate drug candidates that meet the new submission criteria.
